SNAP-8 / Argireline

✦ Cosmetic Research (Lipotec)
SNAP-8 (Acetyl Octapeptide-3) and Argireline (Acetyl Hexapeptide-3)
Also known as: Acetyl Octapeptide-3, Acetyl Hexapeptide-3, Argireline, SNAP-8, topical Botox peptide
Brand names: Argireline (Lipotec), SNAP-8 (Lipotec)

Quick Summary

SNAP-8 and Argireline are related cosmetic peptides that reduce wrinkle depth by partially inhibiting the SNARE complex involved in neurotransmitter vesicle release at the neuromuscular junction. Argireline (Acetyl Hexapeptide-3) is a 6-amino acid fragment; SNAP-8 (Acetyl Octapeptide-3) is its 8-amino acid extension with improved potency.

Skin & Cosmetic Widely Used (Cosmetic)
SNAP-8 and Argireline are related cosmetic peptides that reduce wrinkle depth by partially inhibiting the SNARE complex involved in neurotransmitter vesicle release at the neuromuscular junction. Argireline (Acetyl Hexapeptide-3) is a 6-amino acid fragment; SNAP-8 (Acetyl Octapeptide-3) is its 8-amino acid extension with improved potency. Both mimic the N-terminal end of SNAP-25, competing with the SNARE complex to reduce acetylcholine release at motor nerve terminals, producing localized muscle relaxation topically without the systemic effects of botulinum toxin.

Mechanism of Action

SNARE Complex Competition

The SNARE complex (VAMP, Syntaxin, SNAP-25) mediates vesicle fusion and neurotransmitter release. Argireline and SNAP-8 mimic the N-terminal 6 or 8 residues of SNAP-25, competing for assembly of the SNARE complex and reducing acetylcholine exocytosis at neuromuscular junctions.

Localized Muscle Relaxation

Reduced ACh release produces partial relaxation of facial expression muscles, reducing the repetitive muscle contractions that cause dynamic wrinkles (forehead, periorbital, glabellar).

SNAP-8 vs. Argireline

SNAP-8's two additional amino acids increase SNARE-binding affinity. Studies suggest SNAP-8 achieves similar wrinkle reduction at lower concentrations than Argireline alone.

Research Summary

Argireline Clinical Studies

In a double-blind RCT, 10% Argireline cream applied twice daily for 30 days reduced periorbital wrinkles by 30% on optical profilometry. Effect was localized to application area.

SNAP-8 Studies

SNAP-8 at 5% concentration achieves comparable wrinkle reduction to 10% Argireline in split-face studies, supporting superior concentration-normalized potency.

Research Protocols

Application

Apply 5–10% concentration serum to target wrinkle area twice daily. Most effective on expression-line wrinkles (forehead, crow's feet, frown lines). Effect builds over 4–8 weeks.

Combination

Often combined with Matrixyl (Pal-KTTKS) for dual-mechanism approach: SNAP-8 reduces dynamic wrinkle formation; Matrixyl rebuilds collagen matrix.

Storage & Handling

Follow formulation-specific storage. Most serums: room temperature away from light and heat; some require refrigeration. Raw peptide powder: 2–8°C, dry. Shelf life 2+ years properly stored.
