Mechanism of Action
Procollagen Fragment Mimicry
KTTKS is a fragment of the C-propeptide of procollagen Type I. When collagen is degraded, this fragment is released as a feedback signal to fibroblasts indicating that new collagen synthesis is needed. Palmitoyl-KTTKS delivers this signal artificially, continuously stimulating collagen production even in the absence of active collagen degradation.Palmitoyl Skin Penetration
The 16-carbon palmitic acid chain (C16:0) dramatically increases lipophilicity, enabling the peptide to partition into and traverse the stratum corneum, the primary barrier to topical peptide delivery.ECM Upregulation
Triggered fibroblasts increase synthesis of collagen types I, III, and IV, fibronectin (cell attachment protein), and hyaluronan synthase (HA production), improving skin hydration, elasticity, and structural integrity.Matrixyl 3000 Combination
Pal-GHK (the tripeptide component of Matrixyl 3000) acts via TGF-β stimulation and independently upregulates collagen synthesis, providing an additive effect on matrix production.Research Summary
Clinical Wrinkle Reduction
In a double-blind, randomized, half-face study over 12 weeks, 3% Pal-KTTKS cream reduced wrinkle volume by 17% and wrinkle depth by 13% vs. vehicle control, measured by optical profilometry. Both measurements reached statistical significance.vs. Retinol
Head-to-head comparisons show Pal-KTTKS produces comparable wrinkle reduction to low-concentration retinol with significantly fewer adverse skin effects (redness, dryness, peeling).Collagen Synthesis Studies
Fibroblast culture studies confirm dose-dependent upregulation of collagen I, III, IV mRNA and protein following Pal-KTTKS treatment, with effects measurable within 24–48 hours in vitro.Matrixyl 3000 Data
The Pal-KTTKS + Pal-GHK combination in Matrixyl 3000 shows additive collagen stimulation compared to either component alone in fibroblast models.Calculate your Matrixyl dose Vial strength, BAC water, exact syringe draw in IU. Free, no signup. Open Calc →
Research Protocols
Standard Application
Apply serum or cream containing 3–5% Pal-KTTKS to cleansed skin morning and evening. Allow to absorb before heavier moisturizers or sunscreen.Target Areas
Nasolabial folds, forehead lines, crow's feet, perioral lines. Any area showing dynamic or static wrinkle formation.Assessment
Minimum 8 weeks for measurable outcome. Full ECM remodeling benefits accrue over 12–16 weeks. Continued use is required to maintain benefits, collagen synthesis returns to baseline when use is stopped.Stacking
Commonly combined with SNAP-8 or argireline/" class="wiki-internal-link">Argireline (for expression line reduction) and hyaluronic acid serums. Synergistic with retinol when alternated to avoid irritation.Storage & Handling
Finished formulations: room temperature, away from light and heat. Raw Pal-KTTKS powder: 2–8°C, dry, protected from moisture and light. Shelf life 2+ years properly stored. Palmitoylation makes the peptide relatively stable compared to unmodified peptides.
