Mechanism of Action
NPFF Receptor Signaling
NPVF-derived peptides activate NPFFR1 and NPFFR2, Gi-coupled GPCRs that inhibit adenylate cyclase and reduce cAMP. In the hypothalamus, RFRP/NPVF signaling inhibits GnRH neurons, reducing LH and FSH secretion. This GnRH inhibitory action is analogous to the avian GnIH system and provides an inhibitory counterbalance to kisspeptin activation of GnRH.
Opioid System Modulation
NPVF and related RF-amide peptides modulate opioid signaling by interacting with opioid receptors and by regulating opioid release. NPFF receptors mediate anti-opioid effects that oppose morphine analgesia and contribute to opioid tolerance. This anti-opioid activity is distinct from the reproductive actions and is mediated through different receptor distributions.

Reproductive Axis Inhibition
PreclinicalRFRP-3 and NPVF suppress the HPG axis by inhibiting GnRH neuron firing and reducing LH pulse frequency in rodents and non-human primates. RFRP/NPVF expression varies with photoperiod, energy status, and stress, suggesting it integrates environmental signals to regulate fertility. NPFFR1 antagonists restore LH pulsatility in hypogonadal rodent models.
Stress and Food Intake
PreclinicalNPVF peptides modulate food intake and energy balance through hypothalamic actions. Central RFRP-3 administration increases food intake in rodents under certain metabolic states, and NPVF expression is regulated by metabolic hormones including leptin/" class="wiki-internal-link">leptin and ghrelin/" class="wiki-internal-link">ghrelin. Stress-induced NPVF expression may link chronic stress to reproductive suppression.

Safety Profile
