Mechanism of Action

ACTH(4-7) Core

The Met-Glu-His-Phe core sequence activates alpha-msh/" class="wiki-internal-link">melanocortin receptors (MC4R predominantly) in the CNS, influencing dopamine and serotonin turnover and improving attention and cognitive flexibility.

BDNF Upregulation

Semax and N-Acetyl Semax increase BDNF expression in the hippocampus and prefrontal cortex, supporting long-term potentiation and memory consolidation.

N-Acetylation Effect

The acetyl group blocks N-terminal aminopeptidase cleavage, which is the primary degradation pathway for Semax in nasal tissue. This extends the peptide's residence time in the olfactory mucosa and increases CNS delivery versus unmodified Semax.
