Mechanism of Action

No IGFBP Binding

Full-length IGF-1 is 99% bound to IGF-binding proteins in circulation, which regulate its bioavailability. DES lacks the N-terminal tripeptide that IGFBPs recognize, so it circulates and acts in free form, explaining its dramatically higher potency at the receptor level.

IGF-1R Activation

DES binds the IGF-1 receptor with similar or slightly lower intrinsic affinity than IGF-1, but its free (unbound) state means far more receptor engagement per mole administered.

Localized vs. Systemic

Due to its short half-life, DES acts locally when injected intramuscularly. Unlike IGF-1 LR3 (which circulates systemically for hours), DES primarily affects the injection site tissue, making it the choice for localized muscle research.

In rodent studies, intramuscular IGF-1 DES injection promotes local satellite cell activation and muscle fiber hypertrophy at the injection site with minimal systemic effects.

CNS Activity

IGF-1 DES is the predominant IGF-1 form in the brain (produced locally by neurons and glia). It plays roles in neuronal survival, synaptogenesis, and myelination, distinct from peripherally-acting IGF-1.

Research Protocols

Reconstitution

Critical: IGF-1 DES must be reconstituted with dilute acetic acid (0.1–1% acetic acid or AA solution), not BAC water or sterile water. Alkaline or neutral pH denatures the peptide. Add acetic acid first, then dilute with sterile saline.

Dosing

50–100 µg intramuscularly into target muscle, 1–2x daily. Dose immediately pre- or post-workout for muscle hypertrophy research.

Cycle

4–8 weeks. Longer cycles risk receptor desensitization and IGF-1R downregulation.
