One-Carbon Metabolism
Folate (vitamin B9) is central to one-carbon metabolism, the cellular system that transfers single-carbon units for nucleotide synthesis, methylation reactions, and amino acid interconversion. The active forms of folate (reduced tetrahydrofolates) carry these carbon units in the cell.MOTS-c and AMPK Activation
MOTS-c is a mitochondria-derived peptide that acts as a retrograde signal, translocating to the nucleus and activating AMPK and the folate/methionine cycle. Research suggests MOTS-c increases demand on one-carbon metabolism pathways, particularly the methylene-THF pool used for nucleotide synthesis and the methyl-THF pool used for methylation.The Depletion Hypothesis
During high-dose or extended MOTS-c protocols, increased flux through one-carbon metabolism may deplete active folate faster than dietary intake replenishes. In individuals with MTHFR C677T or A1298C polymorphisms, where folate processing is already reduced - this depletion risk is compounded. Leucovorin bypasses the MTHFR enzyme entirely, providing pre-reduced folinic acid directly usable by the cell.Why Leucovorin and Not Standard Folate
Standard Folate (Folic Acid)
Folic acid is synthetic and must be reduced twice, first to dihydrofolate (DHF) then to tetrahydrofolate (THF) - before cells can use it. This requires the enzyme dihydrofolate reductase (DHFR), which has limited capacity. Unmetabolized folic acid can accumulate in plasma, which has been associated with masking B12 deficiency and possibly interfering with natural killer cell function.Methylfolate (5-MTHF)
The active methylated form of folate. Bypasses DHFR and MTHFR, generally the preferred supplement for MTHFR variants. However, 5-MTHF is committed to the methylation cycle and does not directly support the thymidylate synthesis branch of one-carbon metabolism.Leucovorin (5-Formyl-THF)
A stable, bioavailable, pre-reduced folate that can enter one-carbon metabolism at multiple branch points, supporting both the thymidylate synthesis pool and the methylation pool. Community Protocol
Typical Use
Community protocols vary, but a representative approach:- Timing: 5-10 mg oral leucovorin taken on MOTS-c injection days, or daily throughout the cycle - Dose range: 5 mg/day (low, most conservative) to 25 mg/day (clinical oncology support doses, generally excessive for peptide buffer use) - Form: Oral leucovorin tablets (5 mg, 10 mg, 15 mg, 25 mg strengths), widely available by prescription or through compounding pharmacies - Duration: For the length of the MOTS-c cycle
MTHFR Variants
Users confirmed heterozygous or homozygous for MTHFR C677T or A1298C should treat leucovorin supplementation during MOTS-c as more strongly indicated, not optional.Consult a physician before use. Leucovorin is a prescription pharmaceutical in most jurisdictions. This is for research context only.
